Description:

RNase R은 E. coli 유래 엑소리보뉴클레아제(exoribonuclease)로, 3′ to 5′ exonuclease activity을 나타내며 거의 모든 선형 RNA(linear RNA) 종류를 효율적으로 분해합니다.

이 효소는 원형 RNA(circular RNA), 라리아트 RNA(lariat RNA) 또는 short 3’ overhangs (less than seven nucleotides)를 가진 이중가닥 RNA(double-stranded RNA)는 분해하지 않습니다.

RNase R은 traditional splicing에 의해 생성되는 라리아트 RNA(lariat RNA) 연구뿐만 아니라, back-splicing을 통해 생성되는 circRNA(circular RNA) 연구에도 적합합니다.

세포 또는 RNA 추출물에서 선형 RNA를 제거함으로써, RNase R은 RNA 시퀀싱(RNA-sequencing)을 통한 원형 RNA 종(circular species)의 확인 및 분석을 크게 용이하게 합니다.

• 50 mM Tris-HCl (pH 7.5)
• 100 mM NaCl
• 0.1 mM EDTA
• 0.1% Triton® X-100
• 1 mM DTT
• 50% glycerol

10X RNase R Reaction Buffer:

• 0.2 M Tris-HCl (pH 8.0)
• 1 M KCl
• 1 mM MgCl₂

Unit Definition:
One unit converts 1 μg of poly-r(A) into acid-soluble nucleotides in 10 minutes at 37°C in 20 mM Tris-HCl (pH 8.0), 100 mM KCl and 0.1 mM MgCl₂.
